Preparation of stable colloidal suspensions of superdisintegrants via wet stirred media milling

摘要 Superdisintegrantsarecross-linkedpolymersthatcanbeusedasdispersantsforfastreleaseofdrugnanoparticlesfromnanocompositemicroparticlesduringinvitroandinvivodissolution.Currentlyavailablesuperdisintegrantparticleshaveaveragesizesofapproximately5-130μm,whicharetoobigfordrugnanocompositeapplications.Hence,productionofstablesuperdisintegrantsuspensionswithlessthan5μmparticlesisdesirable.Here,weexplorethepreparationofcolloidalsuspensionsofanionicandnonionicsuperdisintegrantsusingawetstirredmediamillandassesstheirphysicalstability.Sodiumstarchglycolate(SSG)andcrospovidone(CP)wereselectedasrepresentativeanionicandnonionicsuperdisintegrants,andhydroxypropylcellulose(HPC)andsodiumdodecylsulfate(SDS)wereusedasastericstabilizerandawettingagent/stabilizer,respectively.Particlesizing,scanningelectronmicroscopy,andzetapotentialmeasurementswereusedtocharacterizethesuspensions.Colloidalsuperdisintegrantsuspensionswerepreparedreproducibly.Theextensiveparticlebreakagewasattributedtotheswelling-inducedsofteninginwater.SSGsuspensionswerestableevenintheabsenceofstabilizers,whereasCPsuspensionsrequiredHPC-SDSforminimizingparticleaggregation.Thesefindingswereexplainedbythehigherabsolute(negative)zetapotentialofthesuspensionsoftheanionicsuperdisintegrant(SSG)ascomparedwiththoseofthenonionicsuperdisintegrant(CP).
