摘要
Afterpre-cultureandtreatmentofosmosis,cotyledonsofimmaturepeanut(ArachishypogaeaL.)zygoticembryosweretransformedviaparticlebombardmentwithaplasmidcontainingachimerichphgeneconferringresistancetohygromycinandachimericintron-gusgene.Selectionforhygromycinresistantcallusesandsomaticembryoswasinitiatedat10thdpost-bombardmentonmediumcontaining10-25mg/Lhygromycin.Undercontinuousselection,hygromycinresistantplantletswereregeneratedfromsomaticembryosandwererecoveredfromnearly1.6%ofthebombardedcotyledons.ThepresenceandintegrationofforeignDNAinregeneratedhygromycinresistantplantswasconfirmedbyPCR(polymerasechainreaction)fortheintron-gusgeneandbySouthernhybridizationofthehphgene.GUSenzymeactivitywasdetectedinleafletsfromtransgenicplantsbutnotfromcontrol,non-transformedplants.Theproductionoftransgenicplantsaremainlybasedonanewlyimprovedsomaticembryogenesisregenerationsystemdevelopedbyus.
