  • 简介:Fourtestingmethodsusingcationexchangemembrane(CEM),ammoniumacetate,ASI(0.25molL^-1NaHCO3+0.01molL^-1EDTA+0.01molL^-1NH4F)and1.0molL^-1boilingnitricacid,respectively,wereusedtoevaluatesoilavailableK.ThesoilKtestedbyCEMwassignificantlycorrelatedwiththatbytheother(conventional)methods(r^2=0.43^**-0.95^***).ThesoilKtestedbyCEMsaturatedwithNH4HCO3(15minextraction)wasmostcloselycorrelatedwiththatbytheothermethods(r^2=0.60^**-0.95^***),Potassiumavailability,aspredictedbysoiltest,wascomparabletoactualKuptakebycanolaandwheatgrownonthesoilsingrowthchamber.RegressionanalysesshowedthatplantKuptakewasmorecloselycorrelatedwihtKextractedbyCEM(r^2=0.56^**-0.81^***)thanthatbytheconventionalmethods(r^2=0.46^***-0.81^***),mostcolselycorrelatedwiththatbyNH4HCO3-saturatedCEMfor15min(r^2=0.81^***).andworstcorrelatedwiththatbyHNO3(r^2=0.45^**-0.72^***)

  • 简介:Tocloneandconstructtherecombinantplasmidcontainingthemajoroutermembraneprotein(MOMP)geneofChlamydiatrachomatis(C.trachomatis)andtoexpressthefusionproteininE.coliBL21,theMOMPgenewasamphfiedbypolymerasechainreaction(PCR)fromgenomeofC.trachomatisserovarD.ThefragmentwasclonedintotheprokaryoticexpressionvectorpET-22b(+)afterdigestionwithBamHⅠandNotⅠandtransformedintoE.coliXL1-Blue.RecombinantswereselectedbyenzymedigestionandsequencingandtherecombinantplasmidwithMOMPgenewasthentransformedintoE.coliBL21withIPTGtoexpressthetargetgene.TheexpressionrecombinantproteinswerepurifiedbyNi-NTAaffinitychromatography,andidentifiedbySDS-PAGEandWesternblot.Itwasfoundthata1.2kbMOMPgenewasisolated.TheDNAsequenceofMOMPwasfoundtobejustthesameasthesequencepublishedbyGenBank.ArecombinantplasmidcontainingMOMPgenewasconstructedtoexpressthefusionproteinsinE.coli.SDS-PAGEanalysisshowedthattherelativemolecularweightoftherecombinantproteinwasabout47kDathatwasconsistentwiththetheoreticalpredictedvalue,andthespecificityoftheexpressedproteinwasconformedbyWesternblot.ItconcludedthattheMOMPgenecouldbeexpressedintheprokaryoticsystem,bywhichitprovidedthefoundationforthefuturestudiesonthebiologicalactivitiesofC.trachomatisandforthedevelopmentofvaccineagainstthispathogen.

  • 简介:Objective:Thepresentstudyaimedtoevaluatethepossibilityofusingcoherentanti-StokesRamanspectroscopy(CARS)microscopytodeterminethespecificmolecularmorphologyofcholesteatomabydetectingthenaturalvibrationalcontrastofthechemicalbondswithoutanystaining.Materialsandmethods:SpecimensfromthemastoidandtympanicmembranewithandwithoutcholesteatomawereanalyzedusingCARSmicroscopy,two-photonexcitedfluorescence(TPEF)microscopy,andthesecondharmonicgeneration(SHG)microscopy.Results:Incholesteatomatissuesfromthemastoid,astrongresonantsignalat2845cm-1wasobservedbyCARS,whichindicatedthedetectionoftheCH2hydro-carbonlipidbondsthatdonotgeneratevisiblesignalsat2940cm-1suggestiveofCH3bondsinaminoacids.Astrongresonantsignalat2940cm-1appearedinanareaofthesamespecimen,whichalsogeneratedabundantsignalsbyTPEFandSHGmicroscopyat817nm,whichwassuggestiveofcollagen.Inthetympanicmembranespecimenwithcholesteatoma,astrongresonantsignalwithcorrugatedmorphologywasdetected,whichindicatedthepresenceoflipids.AstrongsignalwasdetectedinthetympanicmembranewithchronicotitismediausingTPEF/SHGat817nm,whichindicatedcollagenenrichment.TheCARSandTPEF/SHGimageswereinaccordancewiththehistologyresults.Conclusion:TheseresultssuggesttheneedtodevelopanovelCARSmicroendoscopethatcanbeusedincombinationwithTPEF/SHGtodistinguishcholesteatomafrominflammatorytissues.

  • 简介:ObjectiveTheaimofthisstudyistoinvestigatetheeffectoftransfectedhTERTgeneoncellapoptosisofnewbornratcochlearbasilarmembranecells(CBMCs).MethodsCBMCsisolatedfromnewbornratcochlearweretransfectedusingaplasmidcontaininghumantelomerasereverasetranscriptasegene(pCI-neo-hTERT),andwerescreenedusingG418toobtainstabletransfectedcelllines.Cellapoptosisratewasanalyzedbyflowcytometry.hTERTandapoptosisrelatedgenesexpressionweredetectedbyreversetranscriptasepolymerasechainreaction(RT-PCR).ResultshTERTgeneexpressionwasdetected72hoursaftergenetransfectionintransfectedcells.TheapoptoticrateoftransfectedCBMCssignificantlyreduced.Expressionofapoptosisrelatedgenescorrespondinglychanged.ConclusionTransfectionofhTERTgeneleadstoreducedapoptosisrateinnewbornratCBMCs.andlowerexpressionofapaf1,Caspase3andBCL2intransfectedcellsascomparedtothatofnormalCBMCs.

  • 简介:Theinsectmidgutepitheliumiscomposedofcolumnar,goblet,andregenerativecells.Columnarepithelialcellsarethemostabundantandhavemembraneprotrusionsthatformthebrushbordermembrane(BBM)ontheirapicalside.Theseincreasesurfaceareaavailableforthetransportofnutrients,butalsoprovideopportunitiesforinteractionwithxenobioticssuchaspathogens,toxinsandhostplantallelochemicals.RecentimprovementsinproteomicandbioinfbrmaticstoolsprovidedanopportunitytodeterminetheproteomeoftheT.niBBMinunprecedenteddetail.ThisstudyreportstheidentificationofproteinsfromBBMvesicles(BBMVs)usingsingledimensionpolyacrylamidegelelec?trophoresiscoupledwithmulti-dimensionalproteinidentificationtechnology.Morethan3000proteinswereassociatedwiththeBBMVofwhich697werepredictedtopossesseitherasignalpeptide,atleastonetransmembranedomainoraGPI-anchorsignal.Ofthese,bioinfbrmaticsanalysisandmanualcurationpredictedthat185maybeassociatedwiththeBBMVorepithelialcellplasmamembrane.Thesearediscussedwithrespecttotheirpredictedfunctions,namelydigestion,nutrientuptake,cellsignaling,development,cell-cellinteractions,andotherfunctions.Webelievethistobethemostdetailedproteomicanalysisofthelepidopteranmidgutepitheliummembranetodate,whichwillprovideinformationtobetterunderstandthebiochemical,physiologicalandpathologicalprocessestakingplaceinthelarvalmidgut.

  • 简介:Inthisstudy,apoly(etherblockamide)(Pebax1657)compositemembraneappliedforCO2capturewaspreparedbycoatingPebax1657solutiononpolyacrylonitrile(PAN)ultrafiltrationmembrane.Ethanol/watermixturewasusedasthesolventofPebaxandtheeffectsofethanol/watermassratiosandPebaxconcentrationonthepermeationpropertiesofcompositemembranewerestudied.Toenhancethecompositemembranepermeance,thegutterlayer,madefromreactiveaminosiliconecrosslinkingwithpolydimethylsiloxane(PDMS),wasdesigned.Theinfluenceofcrosslinkingdegreeofthegutterlayeronmembraneperformancewasinvestigated.Asaresult,aPebax/aminoPDMS/PANmultilayermembranewithhexaneresistancewasdeveloped,showingCO2permeanceof350GPUandCO2/N2selectivityover50.Theblendofpolyethyleneglycoldimethylether(PEG-DME)withPebaxascoatingmaterialwasstudiedtofurtherimprovethemembraneperformance.AfterbeingcombinedwithPEG-DMEadditive,CO2permeanceofthefinalPebax-PEG-DME/amino-PDMS/PANcompositemembranereached400GPUabovewithCO2/N2selectivityover65.

  • 简介:Objective:InfraredTympanicThermometer(ITT)isoneofthemostusefulinstrumentsforaccuratelymeasuringtemperature.TheeffectsofearpathologiesonITTmeasurementremainunclear.Thepurposeofthisstudyistodetermineiftympanicmembraneperforation(TMP)affectsITTmeasurementsinadultpatients.Materialandmethods:Atotalof90adultpatientswithmonauralcentralTMPwereenrolledinthisstudy.Patientswerecategorizedintothreesubgroupsaccordingtoperforationsize(1-3mm,4-7mm,and8-10mm).Thetympanictemperaturesoftheaffectedandunaffectedsites,andsubgroupswerecomparedwitheachother.Results:Thisstudycontained54(60%)malesand36(40%)femalesrangingfrom20to58yearsofage(meanage:30.74±9.61years).ThemeantympanictemperatureofthesideaffectedwithTMPwas36.34oC±0.61oC.Themeantympanictemperatureoftheunaffectedsidewithhealthyandintacttympanicmembranewas36.33oC±0.6oC.ThePearsoncorrelationscoreforthetympanictemperaturesandthesizeofTMPwas0.22whichwasnotsignificant(r=-0.12).Conclusion:TMPandperforationsizedonotaffectITTmeasurementsinadultpatients.

  • 简介:Severalcrosslinkedprotonexchangemembraneswithhighprotonconductivitiesandlowmethanolpermeabilitycoefficientswereprepared,basedonthesulfonatedpoly[(4-fluorophenoxy)(phenoxy)]phosphazene(SPFPP)andnewlysynthesizedwatersolublesulfonatedpoly(cyclophosphazene)(SPCP)containingclusteredflexiblependantsulfonicacids.ThestructureofSPCPwascharacterizedbyfouriertransforminfraredspectroscopy(FTIR)and~1HNMRspectra.ThemembranesshowedmoderateprotonconductivitiesandmuchlowermethanolpermeabilitycoefficientswhencomparedtoNafion117.Transmissionelectronmicroscopy(TEM)resultsindicatedthewell-definedphaseseparationbetweenthelocallyanddenselysulfonatedunitsandhydrophobicunits,whichinducedefficientprotonconduction.IncomparisonwithSPFPPmembrane,theprotonconductivities,oxidativestabilitiesandmechanicalpropertiesofcrosslinkedmembranesremarkablywereimproved.TheselectivityvaluesofallthecrosslinkedmembraneswerealsomuchhigherthanthatofNafion117(0.74×10~5S·s/cm~3).TheseresultssuggestedthatthecSPFPP/SPCPmembraneswerepromisingcandidatematerialsforprotonexchangemembraneindirectmethanolfuelcells.

  • 简介:Flowbatterieswithhighenergydensityandlongcyclelifehavebeenpursuedtoadvancetheprogressofenergystorageandgridapplication.Non-aqueousbatterieswithwidevoltagewindowsrepresentapromisingtechnologywithoutthelimitationofwaterelectrolysis,buttheysufferfromlowelectrolyteconcentrationandunsatisfactorybatteryperformance.Here,anon-aqueouslithiumbrominerechargeablebatteryisproposed,whichisbasedonBr2/Br-andLi+/Liasactiveredoxpairs,withfastredoxkineticsandgoodstability.TheLi/Brbatterycombinestheadvantagesofhighoutputvoltage(3.1V),electrolyteconcentration(3.0mol/L),maximumpowerdensity(29.1mW/cm2)andpracticalenergydensity(232.6Wh/kg).Additionally,thebatterydisplaysacolumbicefficiency(CE)of90.0%,avoltageefficiency(VE)of88.0%andanenergyefficiency(EE)of80.0%at1.0mA/cm2aftercontinuouslyrunningformorethan1000cycles,whichisbyfarthelongestcyclelifereportedfornon-aqueousflowbatteries.
