TheyeastHAL1genewasintroducedintoArabidopsisthalianabyAgrobacteriumtumefaciens-mediatedtransformationwithvacuuminfiltrationunderthecontrolofCaMV35Spromoter.Thirty-threeinpidualkanamycinresistantplantswereobtainedfrom75,000seeds.SouthernblottinganalysisindicatedthatHAL1genehadbeenintegratedintoallofthetransgenicplants'genomes.ThecopynumberofHAL1geneintransgenicplantswasmostly1to3bySouthernanalysis.Phenotypesoftransgenicplantshavenodifferenceswithwildtypeplants.Severalsamplesoftransformantswereself-pollinated,andprogeniesfromtransformedandnon-transformedplants(controls)wereevaluatedforsalttoleranceandgeneexpression.MeasurementofconcentrationsofintracellularK+andNa+showedthattransgeniclineswereabletoretainlessNa+thanthatofthecontrolundersaltstress.ResultsfromdifferenttestsindicatedtheexpressionofHAL1genepromotesahigherlevelofsalttoleranceinvivointhetransgenicArabidopsisplants.
